Abstract
Colorectal cancer (CRC) remains an escalating clinical crisis globally, characterized by formidable mortality rates and a desperate need for enhanced therapeutic paradigms. Current clinical interventions are restricted by severe systemic toxicities, non-specific biodistribution patterns, and the rapid emergence of multidrug resistance (MDR) phenotypes within tumor cells.

However, translating conventional oral paroxetine into a viable oncology therapeutic faces physiological bottlenecks, including poor solid-tumor bioaccumulation and volatile pharmacokinetic tracking. To bypass these limitations, this study utilizes biodegradable Polycaprolactone (PCL) matrices to construct nanoscale delivery vehicles designed to protect the payload, control systemic release, and enhance tumor accumulation via the Enhanced Permeability and Retention (EPR) effect.
